The DNA content of the nucleus of the yeast Candida rec. EH 15, Lodderomyces elongisporus CBS 2605, Pichia guillermondii SO809, Pichia guilliermondii fpl-61, and Saccharomyces cerevisiae D10 has been determined by renaturation kinetics. It was possible to make conclusions for the ploidy by measurements of the DNA content per cell and of the percentages of mitochondrial and plasmid DNA, respectively.

From our data we conclude that Candida spec. EH 15 seems to be a diploid or a neuploid yeast containing a high value of mitochondrial DNA per total DNA. The kinetical complexity of the DNA is 8.1 x 109 D. We did not find any evidence for the existence of plasmid DNA.

Lodderomyces elongisporus CBS 2605 in contrast to Candida spec. EH 15 seems to be a haploid strain with a small content of mitochondrial DNA. For the chromosomal DNA we found a relatively small kinetical complexity (7.4 x lo0 D).

Pichia guilliermondii SO809 is a haploid strain while the strain Pichia guilliermondii fpl--61 was found to be diploid. Only small differences exist between the kinetical complexity of the respective DNAs (Pichia guilliermondii SO809 -8.9 x lo0 D; Pichia guilliermondii fpl-61 -9.4 x lo9 D). Both contain likewise a small amount of mitochondrial. DNA.

Saccharomyces cerevisiae D10 -our reference strainshows the same qualities as those documentated in literature. I n this strain we could determine plasmid DNA.
