Facile glycosidic bond cleavage of 2-pyrimidinone2'deoxynucleosides occurs under mildly acidic conditions IuH 3.0) and ambient temueratum. When a 2-twrimidinone nucleoside residue is nresent in a DNA fragment,~h@imly&s results in an aptkinic/apyrimidinic (&sic) site. The incorporation of ZIpyrKdinones into chemically synthesized DNA pmvides a route for the chemical generation of an abasic site at a preselected position in the sequence.
