A sensitive and simple method for the quantitation of oligonucleotide phosphorothioates in biological fluids and tissues is described. This method is based on the extraction of the oligonucleotide from the biological fuids and tissues and immobilization on a nylon membrane. The membrane-bound oligod

โฆ Synopsis
Sequencing of oligodeoxynucleotide phosphorothioate by a modified Sanger method of sequencing is described. The procedure involves ligation of synthetic oligodeoxynucleotide phosphorothioate to an oligodeoxynucleotide, referred to here as "helper oligonucleotide." The helper oligonucleotide has a region which is complementary to (\mathrm{T} 7) primer. By using DNA polymerase and nucleoside triphosphate mixture, 5 '-labeled (\mathrm{T7}) primer is extended onto ligated oligodeoxynucleotide phosphorothioate, which is then analyzed on gel electrophoresis. 1993 Academic Press, Inc.

To understand the regulation of the production of peptide hormones, it is vital to elucidate their biosynthetic pathways. We chose to study a major regulatory peptide, vasoactive intestinal peptide (VIP), a peptide possessing both neurotransmitter and neurohormone actions. To identify the specific p
A simple procedure for the precise assembly of functional DNA sequences from overlapping fragments is described. The fragments to be joined are cloned in tandem in the proper relative orientation into a vector from which single-stranded DNA copies can be obtained. Single-stranded DNA is cut by a res
