The advent of gene targeting technology in mouse embryonic stem cells has revolutionized the study of developmental biology in mammals. This review aims to highlight genetically engineered (knockout) mice characterized primarily by alterations of the bony skeleton. We will examine how information gained from these mutants has contributed to our understanding of the molecular defects affecting skeletal homeostasis and how this information can now be used to provide us with a more effective therapeutic armamentarium for the treatment of metabolic bone diseases, such as osteoporosis.
