Abstract

Following 5‐azacytidine, the incorporation of thymidine into DNA in regenerating rat liver is modified depending on the time of drug administration. When given shortly before or after partial hepatectomy, 5‐azacytidine leads to the inhibition of DNA synthesis, while its administration 16–40 h prior to hepatectomy results in its enhancement in 24‐h regenerating liver. The observed alterations in the synthesis of DNA are in agreement with the formation of thymidine 5′‐triphosphate in cell‐free liver extract. The significance of altered DNA synthesis and cytologic changes as revealed with autoradiography is discussed.
