method is due to the fact that the colored precipitate generated by the NBT/ BCIP (nitro-blue tetrazolium/5-bromo-4-chloro-3-indolyl phosphate) substrate mixture is insoluble in solvents and, therefore, fixed to the membranes. Thus, rehybridization of filters with a second probe is not yet feasible though sometimes highly needed in order to screen a blot with a second probe. Alternative soluble substrates are available [2], but are not as sensitive as the NBT/BCIP mixture. In conclusion, there is an increasing number of reports which demonstrate the various applications of biotinylated DNA probes in medicine [12]. Certainly, nucleic acid hybridization procedures with biotin-labeled probes are very promising diagnostic tools for routine use in clinical medicine (see also [13]).
