Laminin-1, a major basement membrane glycoprotein, promotes tumor cell malignancy. Incubation of B16-F10 melanoma cells with a peptide containing an active sequence in laminin-1, designated AG-73 (leu-glu-val-glu-leu-ser-ile-arg; LQVQLSIR), enhances in vitro adhesion, migration, invasion and gelatin

β¦ Synopsis
Abstract
Liverβcolonizing tumor cells were selected from Bβ16 melanoma by a stepwise procedure: (1) injection of the original cells through the portal circulation; (2) removal of tumor cells from the hepatic tumor nodules for culture in vitro; (3) reinjection of such cells through the portal circulation and repetition of steps 2 and 3 eight times. The predilection of these selected cells to form tumors in the liver was clearly demonstrated following injection of the cells intraβarterially. The accuracy of the injection technique was controlled by monitoring amounts of injected radioactively labelled Sephadex microspheres. This liverβcolonizing preference was not exclusive, but relative.

Past studies have shown that contact between tumor cells and fibroblasts results in stimulation of collagenase production by the fibroblasts. Membrane fractions prepared by differential centrifugation of sonicated B-16 melanoma cells were shown here to contain a collagenase stimulatory factor(s) (CS
