✦ Synopsis


The complexes of platinum are of groat interest in anticancer research and chemotherapy. The investigazkm of the binding properties of the different adducts formed from platinum complexes and DNA bases by liquid c~ separation and the spectroscopic identif'ication of chromatographic fractions frequently requires a large anm~t of analytical material. It is shown that the combination of ion-pair reversed-phase chromatography and infrared spac~ combined with theoretical methods like molecular modelling and normal coordinaee analysis allows the separation attcl ¢hameterizat~ of 7 ethylenediaminepladnungID (Pt(en)) guanosine complexes in PtCen) guanosine reaction mixtures in the micro hinge. The classification of the monofunetional NI, N3, N7 and the bifonctional NT-N! (two species). N7-N3. NT-N7 addluets was confirmed with the quantitative interpretafio~a of the chromatographic peak area in dependence of the diffemm Pt(en)/guanosin¢ ratio and the thermodynamic smbility of the adduet compounds obtained by molecular modelling calculations. Additionally it was distinguished between the N7 and NI/N3 comxlination of the Pt(en) guanosirm complexes by vibrational spectroscopy and normal coordinate analysis.
