✦ Synopsis
Study of the interaction (or interactions) of e-DNP-L-lysine with bovine colostral anti-DNP IgG, using an equilibrium dialysis procedure revealed that the total affinity constant (K,) decreased with an increase in the antibody concentration. However. the curves in Scatchard and log Q' plots were superimposable if the data were corrected for nonspecific binding by normal IgC,. This negative cooperative effect observed with nonspecific IgC, was abrogated on enzymatic removal of Fc'. Isolated Fc' fragments, on the other hand, exhibited the negative cooperative effect on interaction with e-DNP-L-lysine, thus suggesting that the specific binding and hence the K, was independent of antibody concentration.
