Influence of nicotinamide on metabolic status, oxygen consumption, and radiation sensitivity at the cellular level

In some experimental tumor models, nicotinamide-mediated perfusion increases have been low or nonexistent, with no relationship between blood flow changes and measured tumor oxygen levels. Several different mechanisms have been proposed to account for the apparently variable effects of nicotinamide, including modulation of oxygen consumption via perturbation of cellular NAD+/NADH ratios with consequent changes in tumor oxygen consumption. In the present study, we have evaluated the effect of nicotinamide on pyridine nucleotide levels, energy status, oxygen consumption, and radiation sensitivity on LS174t tumor cells in vitro. Nicotinamide at concentrations ranging from 0 to 7.5 mM, and for exposure times ranging from 0.5 to 4 hr does not induce changes in cellular energy or redox (NAD+/NADH and NADP+/NADPH) status, oxygen utilization, and cellular radiosensitivity.
