Influence of native and oxidized proteins on tumour necrosis factor preactivated PMN leukocytes

✦ Synopsis


Activation of PMN leucocytes was monitored by the luminol chemiluminescence (CL) method. PMNs were preincubated with or without tumour necrosis factor (TNF) and stimulated by latex (standard) or latex coated with native or oxidized proteins. As a modifying agent hypochlorite was used. The time course and total yield of CL were analyzed and compared with the standard. The change of charge, loss of the amino group in proteins and formation of aldehydes was observed as a consequence of chlorination. PMNs did not show significant CL after stimulation with protein coated latex unless preincubated with TNF. The enhancement of CL was TNF-dose dependent for the examined proteins and showed a plateau for 40-60 TNF units per 10' PMNs. Chlorination of adsorbed proteins inhibits the PMN leukocyte response and this effect correlates with the loss of amino groups in oxidized proteins. It was also shown that the presence of aldehydes in the protein did not influence the PMN leukocyte response. All the data suggest that PMNs can internalize oxidized material but only after TNF preactivation.
