S u m m a r y

A c o m p a r i s o n was m a d e of t h r e e different ways of p r e p a r i n g leaf m a t e r i a l of Zea mays L. for t h e 'in vivo' e s t i m a t i o n of n i t r a t e reductase activity. Slicing of l a m i n e a e into l -r a m strips gave t h e lowest overall activities a n d e l i m i n a t e d significant d i u r n a l v a r i a t i o n whilst t h e use of i n t a c t lamineae showed m a r k e d d i u r n a l v a r i a t i o n a n d t h e h i g h e s t overall activity.

SHORT COMMUNICATION

This m e a n s t h a t considerable caution has to be exercised in t h e i n t e r p r e t a t i o n of d a t a o b t a i n e d w i t h in vivo assays and t h a t the use of t h e i n t a c t leaf as opposed to leaf s e g m e n t s m a y be a more desirable alternative.
