✦ Synopsis
The polyacrylamide disc electrophoretic technique facilitates the examination of bacterial cell extracts for the presence and characteristics of specific enzymes ( 14). The simplicity of the technique is countered by phenomena which complicate the interpretation thus limiting the value of the results. We report here an improved method for the separation and staining of bacterial dehydrogenases. The method provides means of solubilizing aggregate and membrane-bound enzymes and also of eliminating nonspecific background stain. Examples of application to cultures of Arthrobacter globiformis and Escherichia coli B are given.
