✦ Synopsis

A novel one-step method was developed for entrapment of proteolytic enzymes, namely carborypeptidase B and trypsin, in polymer composite gel based on poly(N-vinyl caprolactam). Natie proteases and enzymes previously stabked by covalent attachment to poiy(vlnylpyrroUdone-co-acrokln) were lmmobked Ln gel% After lmmobiUzaUon, about 90% and 75% of origlnal trypsin and CPB actlvltles, respectlveiy, were retained. The Immobilized enzymes were active wlthin a wide pH range. The optimum temperature of the entrapped enzymes was approrlmately 25OC ldgber than that of tbe sohtble enzymes. The entrapped enzymes were successfully used to obtain human Insulin from recombinant prolnsulin.

Poly(N-vinyl 2-pyrrolidone-g-citric acid) (PVP-g-CA) hydrogels with varying compositions were prepared from ternary mixtures of N-vinyl 2-pyrrolidone-citric acid-water by using 60 Co β₯-rays. The effect of gel composition on the uranyl ions adsorption capacity of PVP-g-CA hydrogels was investigated.
