✦ Synopsis
Most of the tryptic peptides of triosephosphate isomerase have been separated by highperformance liquid chromatography (HPLC) and identified from their amino acid compositions. HPLC and two-dimensional thin-layer methods were compared with respect to reproducibility, recovery of peptides, and background contamination. HPLC was found to be a much more satisfactory method for the separation of tryptic peptides for high-sensitivity structural studies. HPLC is more reproducible, and the recovery of peptides from HPLC is considerably higher than from two-dimensional thin-layer methods. Also, background contamination problems encountered in the case of two-dimensional thin-layer methods are eliminated by the use of HPLC.
