✦ Synopsis
An extraction of nucleotides by the perchloric acid method has been adapted to rat granulation tissue microquantities (a30 mg). Nucleotide extracts are analyzed by strong anionexchange high-performance liquid chromatography. Adenosine S-triphosphate and adenosine S-diphosphate levels can be accurately quantified from the chromatogram with the use of an internal standard, xanthosine 5'-diphosphate, added at the beginning of each extraction. Such a method can be of interest for its potential applications to human tissue microbiopsies without any major traumatism.
