โœฆ Synopsis

Since the introduction in 1990 of a novel ยฏuorogenic substrate for galactose-6-sulphate sulphatase we have used this substrate for prospective prenatal diagnosis in 10 pregnancies at risk for Morquio disease type A. Chorionic villi were analysed in ยฎve cases. The results indicated an affected fetus in one pregnancy which represents the ยฎrst case of ยฎrst-trimester diagnosis of this disorder; heterozygosity was demonstrated in two cases. Following amniocentesis, two affected fetuses and one heterozygote were diagnosed. The results of the present prospective prenatal analyses conยฎrm our previous retrospective studies and demonstrate the reliability and convenience of the 4-methylumbelliferyl substrate.

We report a case of fetal triploidy in which fetal nucleated red blood cells were isolated from the maternal peripheral circulation at 12 weeks' gestation. FISH analysis with X and Y specific probes revealed three hybridization signals for the X chromosomes in 14 cells. The karyotype as established

Fetal karyotyping of trophoblast cells obtained by lavage of the uterine cavity was evaluated on 86 first-tremester irrigation fluid samples. Villus fragments were observed in 72 fluid samples indicating an 83.7 per cent sampling success rate. The amount of villi in these samples ranged from 1 to 32
