โœฆ Synopsis

A principal advance in the production of drug-metabolizing enzymes has been the development of catalytically self-sufficient cytochrome P450 systems, including additional P450-reductase fusion proteins and Escherichia coil and baculovirus coexpression constructs. Continuing work with glutathione transferases has resulted in the identification of important residues by random mutagenesis screening techniques, as well as in the engineering of model Salmonella typhimurium strains for genotoxicity analysis.

Drug metabolizing enzymes are known to be present and active in most extrahepatic tissues. In this review, evidence is presented that expression and activity of several extrahepatic drug-metabolizing enzymes are regulated in unique ways, which may be associated with tissue functions and activities.
