## Abstract The expression of a 30 kD cysteine endoprotease (EPβB) was studied by in situ hybridization and immunomicroscopy to clarify its role in germinating barley grains. At the beginning of germination, EPβB mRNA was expressed in the scutellar epithelium and aleurone cells next to the embryo.
cDNA clones encoding cysteine proteinases from cotyledons of germinated seeds of Vicia sativa L. have been obtained by means of PCR. Degenerate oligonucleotide primers were designed according to conserved amino acid regions of known cysteine proteinases. The deduced amino acid sequences of the cDNA
