✦ Synopsis

During a study of the renal metabolism of exogenous cr-ketoglutarate (+KG)4 in the dog (l), we found that, in our hands, the previously described enzymic assays for a-KG (2, 3) did not yield complete recoveries of a-KG from blood. In particular, perchloric acid, as a protein-precipitating agent for whole blood, gave low recoveries of a-KG. Furthermore, the previous reports (2, 3) had not indicated the number of observations or the variability of the methods used. Therefore, modifications of the analytical procedures were developed and t.he observations are now reported here. Included are data showing that the method(s) used accurately quantify (r-KG concentration in dog blood, plasma, urine, and kidney homogenates, and that this accuracy is not affected by the presence of other metabolites.

The assay of oxaloacetate and \(\alpha\)-ketoglutarate in biological samples is complicated by their chemical instability and low concentrations. We present a quantitative assay for physiological concentrations of these metabolites by isotope dilution gas chromatographymass spectrometry. Samples are

A rapid, sensitive assay for angiotensin-converting enzyme (ACE) inhibitors is described. Biological samples were diluted with methanol to precipitate endogenous ACE and centrifuged. Supernatants were further diluted with 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid buffer, pH 8. Diluted sampl
