β¦ Synopsis
A flow-injection-analysis system has been developed for the determination of essential fatty acids. The enzyme lipoxygenase (EC 1.13.11.12) was immobilized on CNBr-activated Sepharose to form an enzyme column. The oxygen consumption owing to the lipoxygenase-catalyzed conversion from polyunsaturated fatty acid to hydroperoxy fatty acid was monitored by a flow-through cell containing a Clark-type oxygen electrode. Calibration curves for linoleic and cu-hnolenic acid were nearly identical. A linear correlation was observed between current decrease and fatty acid concentration from 0.1 to 1.2 mM.
