A new method for the routine isolation of mutations of spontaneous origin in the yeast, Saccharomyces cerevisiae, is reported. The technique is based on the observation that inositol auxotrophs die when deprived of inositol. However, if macromolecular synthesis is inhibited, most of the cells surviv

β¦ Synopsis
Inositol-requiring mutant,s of strains of Rhodosporidium toruloides were isolated and the frequency of isolation of auxotrophic and catabolic mutants from them was measured after starvation for inositol. Mutants can be isolated a t frequencies up to 31 times greater than those of nintagenized cultures without enrichment.
