The fluorescent glucose analog, 2-(N-(7-nitrobenz-2-oxa-1,3diazol-4-yl)amino)-2-deoxyglucose (2-NBDG), was used to measure rates of glucose uptake by single Escherichia coli cells. When cell populations were exposed to the glucose analog, 2-NBDG was actively transported and accumulated in single cel

β¦ Synopsis
The first steps in E. coli transformation have been studied. It was found that the cells attain the ability of enhanced uptake of donor exogenous DNA upon Ca2+ treatment. At saturating DNA concentrations the cells were capable to take up irreversibly about 6-10(8) dalton of donor DNA per cell (if to assume that all the cells are competent). About 75% of this DNA was found to be attached to the cytoplasmic membrane. Employing Poisson approximation for distribution of the donor-marker DNA molecules on recipient cells it was found that the efficiency of a marker recombination and expression is about 2-10(-5). The first steps in E. coli transformation have been studied. It was found that the cells attain the ability of enhanced uptake of donor 0exogenous

## Abstract DNA transfer directly from cell to cell (conjugation) is common among prokaryotes, particularly Gramβnegative bacteria like Escherichia coli. The phenomenon invariably requires a set of plasmid genes in the DNA donor cell. In addition, E. coli itself makes limited and specific contribut
When transcription of the E. coli trp operon is blocked as a result of inhibition of ribosomal activities by chloramphenicol, nascent trp mRNA but not RNA polymerase molecules seem to detach from the template DNA. The continued association of RNA polymerase with DNA is inferred because RNA synthesis
The transfecting activity of linear lambda DNA is 100 times higher in calcium treated E. coli K12 (lambda i434) than in non-lysogenic strains: the levels of transfection are 1-2.10(7) and 1-2.10(5) infective centers per 1 mug of lambda DNA respectively. The high efficiency of lysogenic cells transfe
