✦ Synopsis

Abstract

A kinetic study has been carried out on the effects of enzyme concentration, protein (human serum albumin) concentration and ionic strength (inorganic and organic sodium salts) on the activity of human placental alkaline phosphatase in solution. The activity is depressed by serum albumin, and the corresponding complex of the protein with the substrate, decreasing its concentration available for catalysis, is responsible for an effect similar to competitive inhibition of the alkaline phosphatase activity. Activating and inhibiting effects were found for inorganic anions, respectively.

Human placental alkaline phosphatase is a membrane-anchored dimeric protein. Unfolding of the enzyme by guanidinium chloride (GdmCl) caused a decrease of the fluorescence intensity and a large red-shifting of the protein fluorescence maximum wavelength from 332 to 346 nm. The fluorescence changes we
