✦ Synopsis


Excellent investigations of the effect of steroidal structure on the gasliquid chromatographic retention t'ime have been carried out by Vanden-Heuvel and Horning (1) and by Brooks and Hanainen :.2). However, these workers either did not examine the estrogens or recorded only the effect of the aromatic A ring on the relative retention time. To accurately identify on gas-liquid chromatography (GLC) the numerous estrogen metabolites isolated in this laboratory, it was necessary to determine the effect of various functional groups on the GLC behavior of the estra-triene2 nucleus.

Methods

The glass GLC columns (6 ft X 0.25 in. diameter) were packcld with Chromosorb W coated with either SE-30 (1.2% w/w) or QF-1 (1.1% w/w) by the filtration method of Fales et ~2. (3). The temperatures employed were 223" for the SE-30 column and 205" for the &F-l column. While operating either column, the flash heater was maintained at 270" and the argon ionization (SrgO) detector at 290". The rate of argon flow through each column was adjusted to 75 ml/min.
