DNA was measured in small portions of rat uterus myometrium by a modification of established procedures. Portions of minced tissue were denatured in TCA and then dissolved in 1~ KOH. Aliquots of the KOH solution were incubated with Pronase before determination of the DNA concentration by means of diaminobenzoic acid fluorescence. With this method it could be shown that the amount of DNA in the rat uterus myometrium increased by a factor of about 2.5 during pregnancy and decreased again to the virgin control levels within 10 days postpartum. This result contradicts previous histological and electron microscopic data which had suggested that very little cell loss occurred during the postpartum involution.
