✦ Synopsis

The aim of this work was to provide a short overview of existing methods for the determination of free drug concentration and protein-drug binding fraction in plasma. Various methods have been described in terms of principles, evaluation of methods, and applications in recent years, with an emphasis on the chromatographic method, i.e. high-performance frontal analysis (HPFA).

A method is described for the evaluation of drug concentrations in plasma and brain from treated rats. The analyte is recovered from plasma or brain homogenate by liquid-liquid extraction and subsequently analyzed by liquid chromatography/tandem mass spectrometry (LC/MS/MS). A simple experimental pr

Reversible attachment to serum proteins plays a significant role in pharmacokinetics and pharmacodynamics, and a clear understanding of this process is fundamental in the development of the rational use of many therapeutics agents. Over the last few years, it has been demonstrated that immobilized h
