Four stereoisomers of nodulisporacid A (1) were synthesized by the concise approach which includes three-component reaction and subsequent one-pot construction of the whole framework. The 1 H NMR comparison of the derivatives (10-12) revealed the absolute configuration of natural 1 to be 4R,4 0 R,6 0 R.
