✦ Synopsis


An enzyme lmmoblllzatlon techmque to measure pemclllm umcentratlon was developed Penrcdlmase IS Immoblhzed by cross-lmkmg with a very fine film of glutaraldehyde, which IS sprayed on to the sensltlve ends of two different pH glass electrodes An extremely short response time of less than 10 s IS guaranteed with electrodes made by this techmque when measurements of pemcdhn are conducted m batch The electrodes show good stabrbty for 12 days when the sensor remams at room temperature As the response ttme of the sensor IS very short, an Immediate determination of pemcdim-V IS possible by indurating this enzyme electrode m a contmuous-flow system, thereby dzspensmg with the need for an on-lme enzyme reactor The pen&m concentration IS calculated as the difference m the potenttometrrc sIgnal obtamed as a peak when the sample flows past the sensor Sample throughputs up to a maxlmum of 200 h-l can been achieved Samples are Injected mto a canter stream consstmg of a buffer of constant pH Three different ~nflgurations of detection cells were destgned and tested with the pcmcdhnase electrode First the perfo~n~ of a magnetIcally stirred flow cell was studled Sub~quently a modified flow-through cell was tested, where the sensor was placed perpendicular to the dtrectton of flow A wail-Jet ceil was used to test the electrode where the flow direction comnded wltb the electrode axis Fmally, the magnetically stirred flow cell was used to detect pemcdlm-V m fermentatton broth samples Keywor& Enzymatic methods, Flow mjectlon, Potentiometry, Enzyme electrodes, Fermentation, Pentcdhn, Pemcllhnase electrode Analyses of samples contammg pemalhns are of interest wth regard to pha~aceutic~ formu-Iatlons, biological flulds and fe~entatlon broth The determmatlon of pemcdlm usmg pH glass electrodes by lmmobdlzmg pemcllhnase on to the
