โœฆ Synopsis


As pointed out by van Es and Bont (1, 2) absorption optics with an electronic scanning system used in connection with the analytical ultracentrifuge have certain advantages for the determination of molecular weights. These authors have already carried out measurements at exactly the meniscus and exactly the bottom, necessary for the application of Archibald's principle. However, the accuracy was only 30%. This low value for the accuracy originated from the optical part of the system.

Calculations (3) and our own observations with respect to the optical system revealed that no sharp image of the cell could be formed in their experimental setup. We have made successful efforts to improve the optical part. Investigations on a representative series of well-known peptides and proteins showed that very satisfactory values for the molecular weights can be obtained with a minimum of arithmetic effort and in a rather short time.
