✦ Synopsis


The enzymic method of Williamson, Mellanby, and Krebs (1) for the determination of D (-) +hydroxybutyrate is not applicable to material containing relatively large quantities of malate because the p-hydroxybutyrate dehydrogenase preparation used could not be completely freed from malate dehydrogenase. A method for the determination of ,8-hydroxybutyrate which can be used in the presence of malate is described in this paper. /3-Hydroxybutyrate is quantitatively converted to acetoacetate in the presence of ,l3-hydroxybutyrate dehydrogenase, pyruvate, lactate dehydrogenase, and NAD, according to the over-all reaction: /3-hydroxybutyrate
