✦ Synopsis
A rapid, accurate, and sensitive method for the determination of leucine specific radioactivity was developed. It is based upon a popular technique in which a second label is introduced by reaction with radioactive fluorodinitrobenzene, allowing a determination of specific radioactivity from the YYH ratio in the isolated derivative. The present method utilized high-performance liquid chromatography to isolate the methyl ester of the dinitrophenyl derivative. Resolution of the methyl-DNP-leucine from the corresponding derivatives of all other amino acids was achieved on small particle silica packings in an elution time of 6 min. Application to the determination of specific radioactivity of other amino acids is possible.
