Synthesis of the sulfone containing lo-membered enediyne derivative 3 and its cycloaromatization in basic conditions via an ailenic sulfone intermediate as well as its DNA cleaving activities are described.

The new class of antitumor antibiotics calicheamicins,l esperamicins,* and dynemicin As have attracted considerable attention due to their very high biological activity as well as their potent DNAcleaving activities.
