This increase in ACBP occurred within 7 days of exposure to the high fat diet (66.5 + 13.1 vs 105.7 + 24.4 pg/ng DNA) and remained elevated after 28 days of high fat feeding (140.5 + 13.7 pg/ng DNA). A similar pattern of change occurred in the kidney but the magnitude of the change was significantly less (25.0 + 4.4 vs 31.9 + 5.0 pg/ng DNA) than in the liver. ACBP in the heart had increased significantly only after 28 days of fat feeding. The ACBP content was determined in subcellular fractions isolated by density-gradient ultracentrifugation. ACBP was widely distributed throughout the cell. The majority of ACBP was located in the cytosol. Significant amounts were also present in microsomes, peroxisomes and mitochondria. Approximately 80% of the mitochondrial ACBP was present in the matrix of this organelle. These data lend further support to the notation that ACBP may be an important "house keeping" protein and as such play a significant role in the regulation of intermediary lipid metabolism.
