✦ Synopsis


A continuous spectrophotometric assay of deoxythymidine (dThd) phosphorylase is reported which monitors conversion of Snitro-2'-deoxyuridine to S-nitrouracil. The reaction is accompanied by a large increase in absorbance (AE = 10') in a region of the visible spectrum (350 nm) which is free from interfering absorbance of components of cell extracts. Kinetic parameters of the phosphorolysis of NO,dUrd are similar to those of the natural substrate, dThd, and NO,dUrd is a very poor substrate for uridine phosphorylase.
