Dinitrophenyl (DNP)-amino acids have been calorimetrically determined by making use of the red color formed by their reaction with cyanide in dimethy~suifoxide or dimethyiacetamide.

The extinction coefficients are over 20,000, making this method more sensitive than previous methods which have used the intrinsic yellow color of DNP-amino acids. Experiments with model compounds have demonstrated that the red cotor is due to the nucleophili~ attack of cyanide on the activated aromatic ring to form a stable anion.
