The properties of radioiodine are reviewed in relation to its use in labeled compounds. Iodine is characterized by a varied spectrum of oxidation states and can form a variety of complex ions and multivalent compounds. The picture of exchange reaction mechanisms is complex, but can be rationalized in terms of stabilization of intermediate complexes formed with negative, neutral, or positive iodine species. The replacement of organometallic groups by electrophilic iodine is discussed as a synthetic tool,
