The 16,17-Double Bond Is Needed for Irreversible Inhibition of Human Cytochrome P450 17α by Abiraterone (17-(3-Pyridyl)androsta-5,16-dien-3β-ol) and Related Steroidal Inhibitors. -Investigation of the structural features required for irreversible binding of steroidal inhibitors, e.g. (I)-(VI), to the human title enzyme shows that the double bond adjacent to the pyridyl residue as in (I)-(IV) is essential and that binding of steroids without double bond, e.g. (V) and (VI), is reversible. Preincubation of abiraterone (I), already selected for clinical trials, and its similarly potent [→(II) and (III)] or less potent [→(IV)] congeners with the enzyme enhances their activity in the treatment of carcinoma of the prostate. -(JARMAN, MICHAEL; BARRIE,
