Synthesis of the Selective D 2 Receptor Agonist PNU-95666E from D-Phenylalanine Using a Sequential Oxidative Cyclization Strategy. -In connection with the development of compound PNU-95666E (VIIb), a potent and highly selective agonist of the dopamine D 2 receptor which may possess potential as a treatment for Parkinson's disease, a synthesis procedure amenable to scale-up is developed. Critical to the success of this route are two oxidative nitrogen annulations to provide the tricyclic ring skeleton. -(ROMERO, A.
