Synthesis of Novel Fluorinated Derivatives of 1,3-Dimethylbarbituric Acid.

-Fluorinated derivatives of 5-benzylbarbituric acid are potentially interesting as anticancer and anti-AIDS agents. They are prepared using the Diels-Alder reaction of difluoro ketene aminal with compound (I), prepared by Knoevenagel reaction of 1,3-dimethylbarbituric acid with the appropriate aryl aldehydes, as the key step. Compounds (III) and (IV) are found to be inactive in screenings for anticancer activity; the remaining compounds are not tested.
