✦ Synopsis

Synthesis of 1,2,3-Thiadiazolo (5,4-d)pyrimidines.

-Condensation of 5-aminothiadiazole-4-carboxamides (I) or (IV) with orthoesters provides the target thiadiazolopyrimidines in moderate to good yield. Attempts to synthesize this heterocyclic system by three-component condensation of readily available 5-aminothiadiazole-4-carboxylate (VI) with aromatic amines and orthoesters succeed only by use of strongly nucleophilic amines. Less nucleophilic aromatic amines yield the corresponding formamidine intermediates (cf. (IX)). -
