Synthesis and Identification of Conformationally Constrained Selective MMP Inhibitors.

-Cyclic thiol based MMP inhibitors like (IV) are discovered. Reduction of the intermediate β-keto-sulfide leads to a 3:1 ratio of alcohols with the equatorial isomer as the major one. The thiols (V) are prepared using the same procedure as for the preparation of the corresponding acyclic thiols. The novel inhibitors retain the potency of their flexible acyclic counterparts. Cyclopentyl analogue (VI) is significantly less active.
