Characterization of the Cytotoxic Activity of Nitric Oxide Generating N-Nitroso Compounds.

-The NO-generating ability of several N-nitroso compounds (I)-(IV) as well as their cytotoxic activity are studied. The greatest NO-generating activity shows the aromatic N-nitrosoureas (I) and nitrosamides (II) while the aliphatic N-nitrosoureas (IV) are inactive due to the stability of the N-NO bond to decomposition under the reaction conditions used. N-nitrosamines (III) also show low NO-generating ability compared to compounds (I) and (II). The best results among the N-nitrosoureas (I) are obtained for derivatives bearing large substituents at the ureido-N3 nitrogen and bearing p-substituents at the aromatic ring such as compounds (Ia), (Id). In general, a correlation is found between NO-generation and cytotoxicity of the studied compounds.
