✦ Synopsis

A New Regio-Defined Synthesis of PMEA.

-The synthesis of PMEA (VIII) is achieved employing regioselective introduction of the phosphonomethoxyethyl moiety to the pyrimidine nucleus (III) followed by subsequent cyclization to form the purine ring system (VI). The overall yield for the synthesis of (VIII) is 35% from the commercially available chloride (I). -
