โœฆ Synopsis

A previous report from this laboratory (Rothenberg et al., 1983a) demonstrated the presence of an Na+/H + exchanger in human epidermoid carcinoma A431 cells. We now characterize surfacederived membrane vesicles from this cell line which contain a functional Na+lH+ exchanger. The Na+/H+ exchanger in A431 vesicles shares a number of characteristics in common with previously described Na+/H+ exchangers including the following: (1) Na+ uptake is stimulated by an outward-directed pH gradient and inhibited by an inwarddirected pH gradient. (2) Na+ uptake is inhibited by arniloride and its analogs and their relative effectiveness is similar in vesicles and A431 cells. (3) The Na+/H+ exchanger uses Na+ o r Li+ as a substrate but not K + or Cs+. (4) H + efflux is stimulated by an inward-directed Na+ gradient and inhibited by t h e amiloride analog 5-Ndimethylamiloride. The Na+/H + exchanger in these membrane vesicles is activated allosterically by low intravesicular pH. The apparent pKa of the activating site is 6.4-6.6, characteristic of the N A + / H + exchanger before activation by mitogens.

Previous studies have documented the activation of Na+/H+ exchange in A431 cells by the addition of epidermal growth factor or serum (Rothenberg et al., 1983b). Here we show that exposure of A4 31 cells to medium of increased osmolarity also leads to activation of Na+/H+ exchange and to an increase

Tea polyphenols are known to inhibit a wide variety of enzymatic activities associated with cell proliferation and tumor progression. The molecular mechanisms of antiproliferation are remained to be elucidated. In this study, we investigated the effects of the major tea polyphenol (-)-epigallocatech

The apparent volume of neutrophils, as measured electronically with the Coulter counter, has been reported to increase upon treatment with chemotactic factors. The occurrence of a volume change was confirmed by forward angle light scattering and by isotopic measurements of intracellular water space

Some epithelial cells have Na ฯฉ /H ฯฉ exchanger (NHE) activity in both apical and basolateral membranes. Amiloride-sensitive NHE-1 is generally identified in the basolateral membrane. The renal cell line, OK7a, targets amiloride-resistant NHE predominantly to the apical membrane. It is controversial

## Abstract Photodynamic therapy (PDT) is a kind of photochemoโ€therapeutic treatment that exerts its effect mainly through the induction of cell death. Distinct types of cell death may be elicited by different PDT regimes. In this study, the mechanisms involved in the death of human epidermoid carc
