✦ Synopsis
Three cases of protein characterization are presented where mixed-mode scanning sequences are executed. The processes involve identification of sites of glycosylation in fetuin, and phosphorylation in ß-casein (based on work by Carr et al) and determination of the protein sequence of cytochrome c. By implementing different scan modes sequentially it is possible to detect post-translational modifications, such as glycosylation or phosphorylation, and to determine the molecular weight and the partial amino acid sequence in a single experiment incorporating liquid chromatography with ionspray, in source, collision-induced dissociation mass spectrometry.
