✦ Synopsis


The physico-chemical properties of apricot kernel proteins before and after detoxification treatments with water and ammonium hydroxide were determined. Water treament shifted the A,,, to 278 nm while ammonium hydroxide has no effect. Fluorescence spectrum showed emission red shift and quenching for both detoxification treatments. Sedimentation velocity pattern of the untreated proteins contained two peaks having S,,., values of 6.8s and 11.1s. Detoxified kernels showed aggregates of 15.4s and 16.9s due to water and ammonium hydroxide treatment, respectively. Gel filtration pattern consisted of three peaks for all samples, meanwhile ion-exchange chromatography showed an additional peak. HPLC, SDS-PAGE and isoelectric focusing-PAGE proved dissociation of the high molecular weight proteins as well as removal of the low molecular weight protein fractions due to detoxification treatments.

Zusammenfassung

Biochemische Untersuchungen iiber einige nicht konventionelle Proteinquellen. 6. Mitt. Physikochemische Eigenschaften von Aprikosenkernproteinen und deren VerCnderungen wghrend der Detoxifizierung Es wurden die physikochemischen Eigenschaften von Aprikosenkernproteinen vor und nach Detoxifizierung mit Wasser und mit Ammoniumhydroxid bestimmt. Wasserbehandlung fuhrte zu einer Verschiebung von i , , , nach 278 nm; die Ammoniumhydroxidbehandlung hatte keinen Effekt. Im Fluoreszenzspektrum waren eine Emissionsrotverschiebung und Fluoreszenzloschung bei beiden Detoxifizierungsbehandlungen zu erkennen. Die Sedimentationsgeschwindigkeitskurve wies zwei Peaks mit S,,~,-Werten von 8,6S und 11,lS auf. Detoxifizierte Kerne zeigten als Folge der Wasser-und der Ammoniumhydroxidbehandlung Aggregate von 15,4S und 16,9S. Die Gelfiltration ergab 3 Peaks fur alle Proben, die Ionenaustauschchromatographic noch cincn zusiitzlichen Peak. HPLC, SDS-PAGE und PAGE-Isoelektrofokkusierung lieBen ebenso die Dissoziation der Proteine hohen Molekulargewichtes wie die Entfernung der Proteinfraktionen niedrigen Molekulargewichtes als Folge der Detoxifizierungsbehandlungen erkennen.

Protein concentrates were prepared from fenugreek seeds by extraction with distilled water, salt solution and alkaline solution. Alkaline solution resulted in the highest extraction yield (82%). The protein concentrate has a good digestibility (over 90°h) as determined by in vitro enzymatic method.
