We developed gas chromatography-mass spectrometry assays for the concentration and mass isotopomer distribution of propionyl-CoA, methylmalonyl-CoA, and succinyl-CoA in tissues. The assays involves perchloric acid extraction of the tissue, spiking the extract with [ 2 H 5 ]propionyl-CoA and [ 2 H 4

✦ Synopsis
We present two techniques for determining the concentration and ({ }^{13} \mathrm{C}) enrichment of acetate in biological fluids. After the sample has been spiked with an internal standard of (\left[2,2,2,{ }^{2} \mathrm{H}_{3}, 1-{ }^{13} \mathrm{C}\right]) acetate, acetate is first enzymatically converted to acetyl-coenzyme (A), which is chemically converted to acetylglycine. The latter is analyzed by gas chromatography-mass spectrometry, either as a methyl ester by positive chemical ionization or as a pentafluorobenzyl ester by negative chemical ionization. The mole percentage enrichment of tissue acetyl-CoA can also be assayed after conversion to acetylglycine pentafluorobenzyl ester. O 1994 Academic Press, Inc.

We present gas chromatographic/mass spectrometric and gas chromatographic/isotope ratio mass spectrometric assays of the 13C enrichment of plasma urea converted to its dimethylaminomethylene derivative. The limits of sensitivity of the two techniques are 0.2% and 0.02%, respectively. The techniques
The 13C mass isotopomer distribution of liver phosphoenolpyruvate (PEP) yields important information on the regulation of gluconeogenesis and the citric acid cycle. A convenient technique is presented for measuring the mass isotopomer distribution of PEP in tissue extracts. The procedure involves re
