In this review, we will briefly summarise the principles, advantages and disadvantages of the various strategies. The main aim is to summarise recent applications of these methods; however, we will focus on specific advances made by this methodology in our understanding of lymphopoiesis and tumorigenesis. Comparing PCR-based and sequence-based approaches, it becomes clear that both strategies will remain invaluable for some years to come.
