β¦ Synopsis
An enzymatic method for the determination of picomol amounts of dCMP in cellfree extracts was developed. The method is based on the quantitative phosphorylation of dCMP to dCTP by a partially purified dCMP-kinase and a pure NDP-kinase.
The dCTP formed is measured using a DNApolymerase assay. With this technique as little as 10 pmoles of dCMP can be measured accurately. The method was applied to the study of dCMP, dCDP, and dCTP pools in resting and growing cells. * Abbreviations and enzymes: NDP-kinase nucleoside diphospho kinase (EC 2.7.4.6); dCMP-kinase (EC 2.7.3.14); DNA-polymerase (EC 2.7.7.7).
